Technical Design Guidelines

Primer Design

• ความยาวแนะนำ: 30–35 bp
• ไพรเมอร์สั้นเกินไปอาจลดความไวและความเร็วในการขยาย
• 5′ ปลายของ reverse primer ต้องมี Modification (เช่น Biotin)
• หลีกเลี่ยง secondary structures
• Amplicon ที่แนะนำ: 150–500 bp

 
Probe Design (Colloidal Gold System)

• ความยาวโพรบ: 46–52 nt (อยู่ระหว่าง forward และ reverse primer)
• 5′ End: ติด Antigen Tag (เช่น FAM)
• กลางสาย: มี dSpacer (THF) เป็นตำแหน่ง recognition สำหรับ nfo enzyme
• 3′ End: ติด Blocking/Modification group (amine, phosphate หรือ C3-Spacer)
