Technical Design Guidelines

Primer Design

• ความยาวแนะนำ: 30–35 nt
• ไพรเมอร์ที่สั้นเกินไปอาจลดความไวและความเร็วในการขยาย
• Downstream primer ต้องมี 5′ modification (เช่น Biotin)
• หลีกเลี่ยง secondary structure
• Amplicon ที่แนะนำ: 150–500 nt

Colloidal Gold Probe Design

• ความยาวโพรบ: 46–52 nt (อยู่ระหว่าง upstream และ downstream primer)
• 5′ End: ติด Antigen marker (เช่น FAM)
• กลางสาย: มี dSpacer (THF) เป็นตำแหน่งจำเพาะสำหรับ nfo recognition
• 3′ End: ติด Blocking group (amine, phosphate หรือ C3-spacer)
